工作原理
PBMC與血液中的其他成分存在密度差異，利用密度在1.077±0.001g/ml之間而且近于等滲的LymphoprepTM溶液作密度梯度離心時(shí)，紅細(xì)胞和粒細(xì)胞由于密度大，故沉于分離液底部。PBMC密度稍低于分離液，故位于分離液界面上，獲得PBMC。LymphoprepTM分離液含有多糖（polysacccharide）,多糖能粘附在細(xì)胞表面，使紅細(xì)胞和粒細(xì)胞聚集下沉，但它對(duì)淋巴細(xì)胞的活性和紅細(xì)胞和粒細(xì)胞進(jìn)一步分離有負(fù)面影響。

注意事項(xiàng)：
1、該分離液僅適用于人體的外周血液，不適用于動(dòng)物。
2、使用該分離液時(shí)，需將待分離的血液用生理鹽水做等體積的稀釋。
3、分離液的保存應(yīng)置于無菌環(huán)境，并避光保存。
4、詳細(xì)操作步驟見產(chǎn)品說明書。
技術(shù)參數(shù)
成分：泛影葡胺鈉(Sodium diatrizoate)：9.1% (w/v)；多糖：5.7% (w/v)
密度：1.077±0.001g/ml
滲透性：290±15mOsm
內(nèi)毒素含量：<1.0EU/ml
分離所得PBMC中紅細(xì)胞的含量：整個(gè)細(xì)胞數(shù)目的3-10%。
Lymphoprep™ is a density gradient medium recommended for the isolation of mononuclear cells from peripheral blood, cord blood, and bone marrow by exploiting differences in cell density. Granulocytes and erythrocytes have a higher density than mononuclear cells and therefore sediment through the Lymphoprep™ layer during centrifugation. Lymphoprep can be substituted for Ficoll-Paque™ without any need to change your existing protocols and is fully compatible with both Sepmate™ and RosetteSep™.
